Actually you've heard Carl and me talk about the purported testosterone boosting effects of sildenafil aka Viagra on theScience Round Up in the past, but Mr. Spitzer and his colleagues from the Greenfield Health Center are the first to prove the potency of the testosterone response in male subjects who received "an optimized dose" (more on that later) of sildenafil, as well as its "direct action on the testes" in a controlled larger-scale trial (Spitzer. 2013).

More than just "spitz" (engl. "horny")

The data Matthew Spitzer et al. analyzed came from the Testosterone and Erectile Dysfunction Trial in the course of which 140 men, aged 40–70 years with erectile dysfunction, low serum total testosterone (<11.4 nmol/L; 330 ng/dL) and/or free testosterone (<173 pmol/L; 50 pg/mL) over received starting doses of
  • 25mg sildenafil, if they were on an alpha-blocker
  • 50mg sildenafil, if they never used sildenafil before, and
  • 100mg sildenafil, if they were viagra veterans
for  3–7 weeks. The men in the 25mg and 50mg groups had the chance to increase the dosage to up to 100mg "on occasion"; or, in other words, when their sexual desire did not match their physical function.

As you can see in Figure 1 the ingestion of a handful of the infamous blue pills lead to significant increases of in total (+3.6 nmol/L = 103 ng/dL; p<0.001) and free (+110 pmol/L = 31.7 pg/mL; p<0.001) testosterone levels and were accompanied by parallel increases in serum DHT (0.17 nmol/L = 4.9 ng/dL; p<0.001) and oestradiol (14 pmol/L = 3.7 pg/mL; p<0.001) and significant suppression of luteinizing hormone (change 1.3 units/L; p=0.003) levels.

The latter, i.e. the reduced luteinizing hormone levels are what actually suggests that we are dealing with a direct effect at the testicular level and not some weird hyptohalamic feedback. A centrally mediated in crease in testosterone production would after all have gone hand in hand with in- not decreases in luteinizing hormone concentration.

Minimal changes in adrenal hormones

With small, but noticeable increases in androstenedione and oestrone increased by 1.3 nmol/L (38 ng/dL; p=0.011) and 10.7 pmol/L (2.9 pg/mL; p=0.012), Spitzer et al. cannot exclude that the sildenafil induced phosphodiesterase-5 inhibition did also affect the adrenal glands - or, to be precise, "late adrenal androgen synthesis" (Spitzer. 2013):

"The adrenal glands are the major site for production of androstenedione, which is converted to oestrone by aromatase. Enzyme 17,20 lyase, which converts 17a-hydroxyprogesterone to androstenedione, is upregulated with sildenafil treatment in rats.

This could explain the increase in serum androstenedione levels in our participants. However, an increase in 17,20-lyase activity should also result in an increase in DHEAS levels, which was not observed. Further investigation is needed to study the effect of sildenafil on adrenal androgens." (Spitzer. 2013)
While the exact mechanism that's responsible for the androstenedione increases must still be determined, it is, as previously mentioned, almost certain that the increase in testosterone levels was mediated by direct effects on the steroidigenic leydig cells in the testes. They are the ones that respond to the luteinizing hormone signalling from the brain have been reported to respond directly to PDE-5 inhibitors in previous studies by Andric et al. from 2007 & 2010, as well as Janjic et al. from 2012.

More sex = more testosterone?

Whether and to which extend the increase in testosterone has also been indirectly promoted by increases in sexual activity, which have been previously reported to lead to increases in testsosterone levels after successful treatment of erectile dysfunction (Aversa. 2013) remains to be seen. The data from the study at hand does at least not support this notion, because a "greater sexual activity did not predict greater change in serum total testosterone" (Spitzer. 2013).

Other studies have associated greater sexual arousal with acute increases in LH and subsequent increases in testosterone production (LaFerla. 1978; Rowland.1987). With decreasing not increasing LH levels being one of the main findings of the study at hand, it is however more or less impossible that the increased sexual desire was the horse, not the cart in the sildenafil-related increases in testosterone levels Spitzer et al. observed in their 140 borderline to full-blown hypogonadal man.

I don't know if you have noticed, but as of late, there has been a definitive testosterone deficiency as far as the topics of the daily SuppVersity news are concerned. So don't click this article away, thoughtlessly, ladies!

50mg that was, as mentioned in the introduction also the dosage Liu et al. gave their sedentary young (aged 21.4±0.3 years, N=8) and middle-aged (aged 49.3±2.4 years, N=8) male participants 12 h before the exercise challenge under supervision of staff personnel. This exercise challenge was a standardized High Intensity Interval Training (HIIT) comprising 5x2min of cycling at 100% VO2max with 1 min rest between each cycling interval. The exercise was perfomed at 9:00AM and in a fasted state. The latter is probably significant, as the hormonal response of the adrenal gland - albeit only that of cortisol (cf. Labsy. 2013) - depends on the time of the day with a 2-3x higher response in the morning compared to the afternoon (cf. Thuma. 1995; Oskis. 2012).

This protocol was meant to shed some light unto the hitherto unknown effects of DHEA supplementation on testosterone levels during the recovery period after a bout of vigorous exercise. Interestingly, enough, you as a Suppersity addict (or soon to be addict ;-) will be aware that DHEA's beneficial effects on recovery in general have actually been proven - even in a scenario of temporary overreachig (=strategic overtaining for a short time period to elicit a maximal training stimulus; seefigure 1 and read all the details in my May 2012 article).

While the dosage in the study at hand was 2x lower than in the Liao study and administered only once, it could still give us some insights into whether or not the decrease in creatine kinase (and thus muscle damage) Liao et al. observed in 2012 (the study was officially published only 1 month ago) was accompanied by an increase in total and/or free testosterone. Aside from the DHEA-S levels and the amount of luteinizing hormone (LH), the latter were the two main study outcomes in the Liu study (seefigures 2 & 3):

If you take a closer look at the data in figure 2 you will realize that there were increases in total testosterone in both groups. Contrary to the very unsurprising increase in DHEA sulfate, the latter were yet only significant in the middle-aged study participants (mean age 49 years).

"Nice, but where's the data on cortisol and what about free test & LH?"

What is not shown in figure 1 are the cortisol levels of the study participants, which were not significantly different at baseline. After the exercise intervention, the levels did yet increase significantly - irrespective of whether or not the participants were young or middle aged or received the placebo or the actual DHEA pills. Intriguingly, the drop that occured after 24h in both age groups was much more pronounced in the middle-aged guys.

Contrary to the total testosterone levels, the amount of free testosterone increased significantly in both, the young and old study participants (see figure 3), it actually goes without saying that
"[...] the free testosterone to cortisol [FT/C] ratio increased significantly during the training recovery for both the middle-aged and young control groups." (Liu. 2013)
Now all that is good news, but what's even better news is that, at least in the short run, these profound elevations in free testosterone did not lead to lower levels of the "testosterone production trigger" luteinizig hormone (LH), which is released by your hypothalamus and tells your testes to produce more sex hormones. Now why is that important? Well, the answer is simple, a reduction in LH is not just a sign of the dreaded HTPA shut-down, it is the shutdown - or I should say the mechanistic cause of being "shut down" (=not producing any or too little sex hormones on your own).

Ok, higher T levels, lower testosterone to cortisol ratio, but what about estrogen?
A note on performance and estrogenic side-effects: An 8-week exercise intervention by Brown et al. which used 50mg DHEA /day combined with 3x total body workouts per week did not produce significant changes in any of the study parameters. Neither beneficial ones, such as greater strength increases or hypertrophy, nornegative ones such as increases in estrogen or deteriorations of the lipid profile (Brown. 1999). In 2010, on the other hand, Ostojic et al. reported that 4 weeks of 100mg DHEA, despite having no effects on the body composition of the 19-21 year-old soccer players who participated in the study, led to statistically significant increases in total testosterone (+37%), DHEA-S (+197%) and estradiol (+21% compared to the subjects in the placebo group; Ostojic. 2010). The overall significance of these results is yet questionable, since both studies were pretty underpowered (N=10 and N=20 for Brown and Ostojic, respectively). It is still noteworthy that the DHEA induced "estrogen dominance", which is one of the most frequenly heardt arguments against DHEA supplementation was not observed in either of the two studies.
If the above is the question that's just praying on your mind, then this is probably not the first article on DHEA administration you've read and what's more, the other ones were probably not published on the SuppVersity. While it is true that a handful of previous studies have in fact shown an increase of estrogen in response to the administration of DHEA, the important ratio of testosterone to estrogen was maintained even in the majority of those studies, where this effect was observed (for more details read the text in the red box on the right).

Regardless of how "real" the danger of elevated estrogen levels may be, there is actually another way more important question to be answered, which pertains to the sustainability of the results. In view of the fact that the DHEA was only administered once, it is way more likely that the unquestionably beneficial effects the scientists observed could be of very transient / short-lived nature. The minimal changes in luteinizing hormone for example could well become significant (and negative) in the long run. After all a +100% increase in free testosterone is nothing your body is going to ignore for weeks. It's more likely that it will slowly bounce back to the former steady state by either reducing the production of testosterone or that of DHEA.

So even if it's unlikely that the small dosage of 50 mg would eventually shut you down, you may well lose the benefits after one months of continuous supplementation.

That being said, you could also argue that the exogenous DHEA has,  just as TRT or performance enhancing drugs, the advantage of not being subject to diet-, training- and obviously age-induced reductions in their endogenous production. If any of the latter reminds of a situation / period of your life you are in, your chances of seeing benefits would at least be higher - whether they would be high enough to actually manifest in beneficial training outcomes remains questionable, though.


It is unfortunate that science gave up on DHEA, when it became clear that it's not the fountain of youth. I mean, greater fat-inhibition effects than testosterone are nice, no (learn moreabout those)?
Bottom line: The new information the Liu study has to provide does actually go hand in hand with what the data from the Liao study already suggested. The purported "fountain of youth" hormone has potent ergogenic effects in training scenarios, which focus on vigorous physical activity.

Unfortunately, the evidence from these short-term interventions does not tell us anything about the beneficial and detrimental effect the low and high dose protocols from the Liu and the Liaostudies will have. Personally I would assume that the benefits will wane off, but in the absence of corresponding studies, this is mere guesswork. After all, 99% of the previous research on DHEA has been done in sedentary old men and women. Few trials investigated the effects on young or middle aged subjects and less than a handful did that in an exercise scenario with any resemblances to yours or mine.

So there is probably need for at least another SuppVersityarticle (obviously based on a novel study), before you will hear me recommend the use of DHEA as an ergogenic supplement. New Study Says: Boron Does Indeed Raise Free Testosterone

There have been rumors on the Internet for quite some time - "Did you hear? Boron increases testosterone!" Hitherto, however, these claims have not been substantiated by reliable scientific research. A recent study done by a group of scientists from Iran (Naghii. 2010) yet seems to prove that this is more than the steroid-loaden phantasy of an average gymrat.

In a placebo controlled study, the scientists found that supplementation with 10mg Boron/day did not only lower SHGB and thus raise testosterone, it did also lower three inflammatory biomarkers(Interleukin-6 (IL-6), high sensitive CRP (hsCRP) and tumor necrosis factor-alpha (TNF-α)):

As you can see in table 1, the testosterone to estrogen ratio more than doubled (0.31 to 0.67). This alone makes me believe that it won't take too long for supp companies to notice this little trace mineral and to include it in more and more of their products. Hitherto it is already found in some formulations, like multi-vitamins, meal replacements and other stuff that is enriched with minerals the dosages, however, are minuscule and far from the 10mg used in the present study.

The scientists also investigated the time-course of the effect of boron supplementation on day 1 of the study and interestingly it took only 6 hours for free testosterone to rise from 8.55pg/ml to 11.25pg/ml. To me, this suggests that there is more to boron supplementation than just repleting exhausted blood and/or tissue levels.

Update: I have been digging a little into the possible mechanism of action behind the increase in testosterone and my personal take it that boron is a (weak) anti-estrogen. By cutting estrogen by 50% (cf. table 1) your body will automatically reduce SHGB, which is up-regulated by estrogen. This frees testosterone and may in fact provide (minor) benefits in muscle gain, libido & fat loss. I suspect that in the end this could be related to a reduction in manganese and an increase in magnesium & calcium retention (cf. Acu-Cell information about boron) that follows the administration of large amounts of boron. These is highly speculative and may only be part of the picture, though.
Also, in view of conflicting evidence from previous studies (e.g. Nagghi. 2006), which found that supplemental boron increased (not decreased) estrogen in men, further investigations are warranted.

As the data in figure 1 goes to show these changes were unfortunately transient and the impressive +189% increase in the testosterone to estrogen ratio which occurred during the first month of treatment totally disappeared within the next four weeks. On the other hand, the effects on SHBG the scientists had expected based on the assumption that both SHBG and caffeine intake have been found to be associated with lower risk of type II diabetes in large epidemiological studies, was non-existent in the first and second 4 weeks of the study... at least in the male subjects who were all overweight, nonsmokers and habitually coffee consumers, who had been required to abstain from caffeine intake for at least 2 weeks before the study was conducted.

If you take a look at the data from the female participants (likewise overweight non-smokers, habitual caffeine consumers and, at the beginning of the study, 'dried out'; see figure 2), a different image emerges, in the women we do in fact see a transient rise in SHBG, which goes hand in hand with a decrease in testosterone, de to which the T/E ratio drops by -36% and -54% in the groups drinking caffeinated and noncaffeinated coffee respectively. Just as in their male counterparts, the levels did go back up in the second part of the study so that all values, including the SHBG levels were back in to normal after 8 weeks (please note changes in the 20% range are irrelevant and could be due to having a meal before the test, bad sleep, whatever).

The relative data tells only part of the story

If we do now take a closer look at the actual data and discard the comparison to the control group the scientists the picture becomes even more complex. After all the data in figure 4clearly indicates that we are dealing with a combined effect here. It is correct that the ingestion of the caffeinated beverage had pronounced effects on the T/E ratio especially in the male participants, what the data in figure 1 does yet not tell you is the fact that this effect was also so pronounced, because simply stopping to drink caffeine reduced the T/E ratio from 18.1 to 8.4, i.e. by 54%(!).

Now this certainly reduces the effect size, but it does not totally negate the effect. After all the 'real' coffee drinkers (w/ caffeine) did still increase their T/E ratio from 16.2 to 24.2 -- a certainly likewise noteworthy increase of +29% that is however still far away from the exorbitant +189% increase compared to the poor guys who did not just lose their coffee, but also their virility.

So what does this tell use? 

How cares about SHBG anyways? You should! After all there is relatively conclusive evidence that normal (not exorbitantly high!) SHGB levels have aprotective effect against breast cancer in women and mechanistic evidence that they increase the risk of prostate cancer in men. In both cases SHBG acts independently via the largely ignored SHBG receptor that modulates the action of estrogens. Co-activation of SHBG and estrogen receptor in the prostate induces similar effect on prostate specific antigen secretion as DHT (Nakhla. 1997). Since estrogen alone does not have this effect, it is no wonder that stinging nettle root (Urtica dioica), with its SHGB inhibiting effect is a viable tool in the treatment of benign prostatic hyperplasia (Hryb. 1995). In the female breast, on the other hand, SHBG seems to " trigger a 'biologic' anti-estrogenic pathway" (Fortunati. 1999) and does therefore exert anti-instead of pro-carcinogenic effects.
I guess there are more than just the following three lessons to learn from this study, but at the moment these appear to be the most important ones for me:
  1. The beneficial effects habitual coffee intake has on type II diabetes risk are, contrary to the scientists hypothesis, not mediated by its effect on SHBG.
  2. In overweight men caffeine has a very shortlived beneficial effect on testosterone and the testosterone to estrogen ratio. After 4 weeks the levels do yet return to baseline, so this cannot explain the long-term benefits of habitual caffeine consumption either (maybe you should cycle caffeine instead of testosterone booster < I am just kidding ;-).
  3. In overweight women, there is a similar, yet negative effect on testosterone levels, which is likewise transient and lasts less than 8 weeks. 
  4. The caffeine ads to the 'pro-testosterone' effects, but even decaffeinated coffee has some effects.
  5. Stopping "cold turkey" is not a good idea, when you are "on caffeine"
Now, what is important here is that we are dealing with overweight individuals, in whom the endocrine millieu is usually off. In particular, men tend to have reduced, women tend to have increased androgen levels (think PCOS). The effects we see after short-term withdrawal and the subsequent consumption of a non-negligible amount of 5 cups of coffee everyday could actually have corrective effects on the endocrine milieu, of which both, men and women could benefit, if they would last for more than 4-6 weeks. The detrimental effects of stopping, on the other hand, could be due to the sudden absence of the benefits of caffeine.

Regardless of whether you stop or start drinking coffeine, the endocrine "disturbances" are relatively short-lived and only further testimony to the fact that our bodies will always try to find a new "steady state" in what they consider normal.

Bottom line: There are a myriad of good reasons to drink coffee, getting more manly or more feminine is yet not one of them. Disappointed? Well, on the other hand this means coffee is no endocrine disruptor - and at least in this overweight population it seems to have a marginally beneficial baseline effects (thus the detrimental effects of abstinence).

True or False - High or Low Protein Intakes Have Profound Influence on Testosterone, SHBG, Estrogen, Cortisol & Co?

We are what we eat! Acknowledged, but does this also go for your hormones and different in protein intakes? Let's have another look at the contemporarily available research to figure that out.
In recent study in the International Journal of Sport Nutrition and Exercise Metabolism a group of researchers from theFaculty of Physical Education and Recreation at theUniversity of Alberta reports: "Supplementing a typical daily food intake consisting of 0.8 g of protein·kg-1·d-1 with a wheyprotein isolate (an additional 0.8 or 1.6 g·kg-1·d-1) [...] had no effect on glucose, insulin, testosterone, cortisol, or growth hormone following the final meal" (Forbes. 2013).

I know, anything else would have been a real shocker. What it wouldn't be, though, is a total surprise. If you dig through the available literature you can easily find data that would support the scientists' assumption that doing an experiment "to investigate the effects of a controlled typical one day diet supplemented with two different doses of whey protein isolate on blood amino acid profiles and hormonal concentrations following the final meal" (Forbes. 2013) would be a good idea.

True or false? Certainly TRUE!

No, I am not kidding you! We do have plenty of evidence of direct interactions between chronic high vs. low protein intake and the production and metabolism of hormones, like testosterone, cortisol, their corresponding binding proteins, SHGB and CBP, and a whole host of other molecules that act like hormones, although we often don't call them "hormones". Examples? Well, here you go:
  • Low protein = low SHBG (Longcope. 2000) - At least in the 1552 men in their "best age" (40-70 years) who participated in a study by scientsts from the University of Massachusetts Medical School the consumption of a diet that had less than the average 80g/day of protein in it lead to decreases in SHBG and corresponding increases in free (~bioavailable) testosterone. Age, dietary fiber, and smoking, on the other hand were positively correlated with SHBG.
  • Chronically high protein intakes (40% of total energy) lower testosterone / cortisol ratio (Oi. 2001) - The highly significant increase in cortisol and corresponding decrease in the testosterone to cortisol ratio, Oi et al. report in a 2001 paper have luckily (a) been observed in rodents, only, and that's actually the main finding of the study (b) could be countered by the administration of garlic extract.

    I can only speculate whether and to which degree the endocrine effects of high protein diets differ between rats and men (the Anderson study discussed below suggests they don't) - and you know I love facts and hate speculations and unsupported hypothesis. So I stick to an assumption I could support, i.e. the supposition that the improvements in testosterone, the researchers observed, when they fed their rodents garlic was mediated  by its glutathione (GSH) boosting effects. These effects - and you as a regular SuppVersity reader know that are not "garlic exclusive". Whey protein, for example will also increase GSH (Bounous. 1989). Obviously whey doesn't increase your testosterone, but a "protein overload" from a glutathione boosting protein source may not have to be countered by GSH boosters in the first place.
  • A high protein : carb ratio decreases total testosterone levels in man (Anderson. 1987) - Some people live by the principle that things that mustn't be. As a SuppVersityreader you obviously don't belong to this group of people and will thus be willing to accept that Anderson et al. were able to show that...
    "[...] the testosterone concentrations in seven normal men were consistently higher after ten days on a high carbohydrate diet (468 ± 34 ng/dl, mean ± S.E.) than during a high protein diet (371 ± 23 ng/d1, p<0.05) and were accompanied by parallel changes in sex hormone binding globulin (32.5 ± 2.8 nmol/1 vs. 23.4 ± 1.6 nmol/1 respectively, p<0.01)." (Anderson. 1987)
    As a SuppVersity reader you are yet also smart enough to know that this data is irrelevant, without adequately measured free testosterone levels: If we do the math and calculate the latter (obviously not 100% accurately), i.e. free testosterone on normal protein diet: 9.4 ng/dL  =  2.01 % vs. free testosterone on high protein + low carb diet: 9.02 ng/dL  =  2.43 %, the difference does no longer look so bad, anyway - does it?

    This would not change the fact that you'd suffered a -42% decrease of the testosterone : cortisol ratio from 60.5 to 35 which occurred in the Anderson study, when the participants were put on a meat, fish, poultry, egg white, and protein supplement based 44% protein, 35% carbohydrates, 21% fat diet.
  • A high protein very low calorie diet can increase testosterone levels, but it will do so only if it is used to produce significant weight loss in obese adolescents, whose abundant body fat stores are gnawing away their androgens. In a corresponding study by Brown et al., the eight 11-15-year-old boys and girls where put on liquid (starvation) diets with a protein / carb / fat ratio of 67% / 28% / 5% that containing a total of 492-709 kcal per day.

    As you can see in Figure 3 the significant reduction in body weight (13.5 kg total, 70-75% fat) went hand in hand with improvements in insulin sensitivity and the previously mentioned increase in total testosterone levels in the 14 year-old subjects 3 & 7 and the 18 year-old male adolescents (subject 6) - the only subjects for whom a complete testosterone panel was available.
  • High protein diets are driving forces of GH induced skeletal muscle IGF-1 expression (Sanchez-Gomez. 1999) - With IGF-1 being equally important to men and women, it does not matter that the "subjects" in the study at hand were female growing rats that received either a high- or a low-protein diet with crude protein contents of 222 and 83 g/kg respectively.

    After 14 days on which the rodents were concamittantly injected with saline control, or growth hormone (rhGH) or recombinant human IGF-I (rhIGF-I) at dosages of 350 and 500µg/day, respectively, Sanchez-Gomes et al. observed that
    • Learn more about IGF-1.
      ...the low-protein diet alone reduced IGF-I concentrations in serum and in tissue taken from the gastrocnemius muscle as well as IGF-I mRNA from the same muscle, significantly.
    • ...the high protein diet amplified the retention of injected rhIGF-I in the muscle tissues and was associated with significant improvements of the nitrogen balance
    Based on these results, Myriam Sanchez-Gomez conclude that "the level of dietary protein ingested regulates not only the effect of IGF-I on whole-body N economy but also the regulation of IGF-I gene expression in muscles" (Myriam Sanchez-Gomez. 1999). 
  • Increased f 2-hydroxylation of endogenous estrogen with high protein diets could have cancer protective effects (Anderson. 1984) - Certainly far fetched but not impossible is the connection between the increased 2-hydroxylation of endogenous estrogen in response to high protein diets Anderson et al. observed in a 1984 study in male study participants and the significant association between lower ratios an the risk of breast cancer (Liehr. 1996).

    It goes without saying that this is nothing but a hypothesis - a hypothesis that would yet be supported by a 1999 paper on "dietary predictors of breast cancer recurrence and death" by Saxe et al. which is based on one of the few papers that did not control for "meat" intake (you know that Pizza Salami qualifies as "meat", right?) in which each 5% increase in protein intake was associated with risk reductions of 18% for breast cancer recurrence and 44% for death in premenopausal women (Saxe 1999).

    And what's more, the fact that the associations were less pronounced in the post-menopausal study participants (the "low estrogen counterpart", if you will; see Table 1) only support the notion that the protective effects of protein may (at least in part) be mediated by its effects on estrogen metabolism.

Vitamin D Supplementation(!) Increases Testosterone in Men

Thanks to Dashforce from the MindAndMuscle-Forum we finally have evidence for the direct effect ofvitamin D3 supplementation on testosterone in men. Dashforce reports on a study by scientists from Austria and Germany (Pilz. 2010) who provided 54 men, whose serum levels were previously at the lower range ~30ng/ml (the referential  minimum varies depending on which source you cite from 30-50ng/ml),with either 83 μg (3 332 IU) vitamin D daily or placebo for 1 year.

As you can see in table 1,vitamin D supplementation lead to a significant increase in total testosterone from 10.7 nmol/l to 13.4 nmol/l. What's even more important, this increase (+25%) was not compensated by and an increase of SHGB, so that the amount of free testosterone rose from 0.222 n/mol to 0.267 nmol/l, i.e. by +20% (vs. +5% in the placebo group).

What is particularly interesting about this study is the fact that supplementation actually raised the level of testosterone. This is different from most previous studies, which observed a correlation between higher vitamin D levels and higher testosterone levels. If for example some probands would simply have been more healthy and thus exhibit higher levels of both, we would have had the same correlation. In the case of the Pilz study there is a clear causative effect between restauration of adequate vitamin D levels and increases in total and free testosterone.

Willoughby and Leutholtz assigned their participants in a random, double-blinded fashion to one of the two study arms. Subjects in both arms of the study followed a standardized periodized 28-day resistance training program split into 2 upper-extremity (A1, A2) and 2 lower-extremity (B1, B2) exercise sessions each week. With an overall training volume of 16 exercises sessions with 9x upper- and 8x lower-body exercises
  • A1, A2 - upper body: bench press, lat pull, shoulder press, seated rows, shoulder shrugs, chest flies, biceps curl, triceps press down, and abdominal curls
  • B1, B2 - lower body: leg press, or squat, back extension, step ups, leg curls, leg extension,heel raises, and abdominal crunches

The participants performed 3 sets of 10 repetitions at 70% - 80% of the previously established 1-RM for all exercises and had 2 minutes of passive rest between the sets. Contrary to the said VPX studies, the participants had to refrain from taking any kitchen-sink pre-/post-workout supplements, as well as the obvious roids, prohormones or other ergogenic substances that would compromise the study results.   • 4 capsules containing 3 g of guar gum (PLA), or
  • 4 capsules containing 3 g of D-ASP (Better Body Sports, Ventura, CA, USA)
The dosing was identical to the manufacturers recommendation and based on the previous study by Topo et al. (2009) that caused the whole DAA hype back ~5 years ago (see figure 2, as well).

"Based on our data presented herein, this [=the fact that the serum levels of DAA were higher than normal, but nothing happened] may indicate another potential mechanistic reason why the HPG [hypothalamus > pituitary > gonads] axis was not affected by D-ASP supplementation. Although we observed nonsignificant increases in serum D-ASP in the DAA group, we showed significant increases in DDO levels in response to D-ASP supplementation. The degradative role of DDO is to catalyze the oxidative deamination of D-amino acids to generate the corresponding 2-oxo acids, along with hydrogen peroxide and ammonia (or methylamine).

In rodents, the administration of D-ASP was shown to increase DDO activity (Nagasaki. 1994; Yamada.1989), suggesting that DDO activity is induced by increased levels of D-ASP. Based on this information, in the present study, it is possible that because of the higher baseline levels of testosterone, as a means of androgen-regulated feedback of the HPG axis, the level of serum D-ASP induced by supplementation was conceivably being degraded by DDO at a rate that rendered it unable to effectively activate the HPG axis." (Willoughy. 2013)
Or to put is simply: If DAA works at all, it works only in guys like the participants of the 2009 study by Topo et al. who had baseline testosterone levels at the lower end of the normal range (4.5ng/ml) at the beginning of the study and average levels (6.4ng/ml) at the end of the study period. In trained athletes with testosterone levels in the range of 8ng/ml such as the guys in the Willoughby study the negative feedback will prevent any further increase in testosterone.

  In 1984 I was asked to write an article about ergogenic aids for a national sports magazine. It was to be included in a special issue covering the Summer Olympic Games that year. I interviewed a number ofexperts. One, a medical doctor, told me off the record that he had about 20 Olympic athletes on a regimen that included anabolic steroids and growth hormone. Significantly, none of the athletes got busted during the Olympics. While the guy was forthcoming—although he refused to identify the athletes using the drugs—other experts proved far more secretive.

  I called a man in Boston who was described to me as a “well-known physician.” He had extensive experience with a little-known substance calledSmilax officinalis, which I understood was some kind of herbal concoction, but when I called “Dr. X”—he refused to divulge his real name—he told me that he wasn’t at liberty to provide any details about Smilax. It wasn’t until later that I found out that Smilax officinaliswas actually sarsaparilla, a common flavoring in root beer. A search for the anabolic effects of the plant turned up one obscure connection related to natural compounds called plant sterols.

  Flashback to 1942. A renegade chemist from the University of Pennsylvania named Russell Marker was acting like Indiana Jones, searching for plants with special properties in the wilds of Mexico. At the time, steroid hormones, including testosterone, were being produced in a very expensive process. Working in his lab in 1936, Marker successfully developed a method of producing chemical steroids from plant sterols. All steroids are made from the parent compound, cholesterol. When Marker noticed that plant sterols, which are the plant versions of cholesterol, have a close structural similarity to cholesterol, he was able to produce a human steroid from sarsaparilla. It was too expensive, though, to be used in mass quantities; hence Marker’s Mexican trek.

  Marker found his ideal substitute in the Mexican yam. Diosgenin, a plant sterol in the yam, proved to be an effective substrate for steroid production. Through a five-step process involving the use of various enzymes, Marker successfully produced the steroid progesterone. The first major steroid produced from cholesterol, progesterone is the precursor of other steroid hormones, including estrogen and testosterone.

  Marker offered his discovery to drug companies, but they all turned him down except for a small, little-known company called Syntex. Syntex eventually became a major producer of hormone drugs, including a number of anabolic steroids. Meanwhile, the Mexican yam and diosgenin became the major raw materials for steroid hormones, particularly birth control pills, until they were supplanted by a synthetic process in 1970.

  Mexican yams were also sold as a food supplement because of their plant sterol content. In fact, early forms of supplemental DHEA primarily contained Mexican yam extract. That proved problematic, since the yam itself doesn’t contain any active steroid hormones, including DHEA. The poor quality and ineffectiveness of the supplements led to their removal from the market in 1987 by the Food and Drug Administration. Real DHEA turned up a few years later, thanks to the Dietary Supplement Health and Education Act of 1994.

  It’s no secret that anabolic steroids help build muscle size and strength. The problem with steroids is twofold. First, they’re prescription drugs and have potentially serious side effects. Second, most doctors are loath to prescribe them, knowing how easily they can be abused by those seeking to build muscle. That led to the pro-hormone industry, which began with legitimate DHEA supplements and ended with the advent of the Anabolic Steroid Drug Amendment in 2005.

  Not much better than DHEA, the initial pro-hormones were far worse from a health perspective. They offered little in the way of true anabolic activity but plenty in the way of anabolic-steroid-related side effects, including rapid conversion into estrogen. The last generation of pro-hormones was far improved, since many were actual anabolic steroids that were never released by the companies that researched them—and for good reason. Most of them had too many side effects, such as being hard on the liver. Even worse, evidence for significant anabolic effect in humans was lacking.

  After the FDA banned pro-hormone supplements, supplement companies catering to bodybuilders and athletes were temporarily at a loss. What could they do to maintain their lucrative sales? A few threw all caution to the wind by attempting to sell a few more rejected steroid compounds, while others began to surreptitiously include a new category of steroids, called “designer steroids,” since they were supposedly not detectable by the usual drug screens. That proved short-lived, for the FDA was on the lookout for any substance that could directly convert into testosterone, which put it in direct violation of the 2005 steroid amendment.

  Now, how to explain the current plethora of supplements advertised as boosting testosterone? What the supplement companies did amounted to going back in time. Most of the products are based on plant compounds, which makes them completely legal in the eyes of the FDA. The more important question is, Are they effective?

  Some of the more unscrupulous companies resorted to highly questionable tactics. For example, many current pro-hormone supplements are based on DHEA, but it’s usually called something else. That’s a common tactic today—listing complex chemical names to make consumers believe that a supplement contains a new and revolutionary compound. Another likely motive is that the chemical nomenclature imparts a druglike aura to the product. DHEA is not junk; it benefits those whose bodies produce low amounts of it. As a testosterone-booster, though, it’s effective only for women.

  Plants don’t actually contain real testosterone but rather plant sterols. The human body lacks the enzymes required to convert plant sterols into true steroids, although it can be done in a lab. That explains why plant sterols were—and in some cases still are—used to produce steroid drugs. One plant known to contain an actual androgen—the truffle—is a type of mushroom. Truffles naturally contain a very weak androgen called androst-16-en-3-ol. It isn’t very anabolic, which explains why you don’t see any mushrooms competing for the Mr. Olympia title.

  Perhaps the foremost herbal testosterone booster is Tribulus terrestris, which grows wild in Bulgaria and elsewhere. Tribulus contains various plant sterols, including diosgenin. Most of the sterols are categorized as saponins, since they produce a soaplike effect when metabolized. The active anabolic ingredient in tribulus is a saponin called protodioscin.

  Purveyors of tribulus often note that if it doesn’t contain enough protodioscin, it’s useless for building muscle, although it does have other properties, including diuretic effects and coronary artery dilation. Normally tribulus contains about 5 percent protodioscin. One study analyzed commercial tribulus supplements for their protodioscin content. It varied, with samples from Bulgarian sources having more and those from China and India the least. Quality control is a real problem with tribulus, as it is with many other herbal supplements.

  Tribulus came to the attention of Western athletes after poorly controlled Russian research revealed that it increased testosterone. The initial studies involved animals, such as rams. In the human studies that followed, tribulus was given to men with clinically low testosterone levels. One study found that men receiving 750 milligrams of tribulus daily for five days showed a 72 percent increase in luteinizing hormone, along with a 40 percent rise in testosterone. Less publicized was the accompanying 81 percent rise in estrogen.

  Luteinizing hormone is produced in the pituitary gland and governs the production of testosterone in the testes. It’s suggested that tribulus contains a substance called harmine that blunts the breakdown of an enzyme in the brain that degrades dopamine. Elevated dopamine encourages the production of not only luteinizing hormone and subsequent testosterone but also growth hormone. The much touted “active” tribulus ingredient, protodioscin, aids DHEA synthesis and release.

  Based on these mechanisms, tribulus should indeed be an efficient testosterone booster, although increasing DHEA wouldn’t have much of an anabolic effect in young men. Animal studies seem to confirm the hormone-boosting effect of tribulus. One recent study, in which primates, rats and rabbits were given a tribulus extract intravenously, resulted in a 52 percent increase in testosterone, a 31 percent rise in the testosterone metabolite DHT and a 29 percent rise in DHEA-S in the primates. The rats in the study were castrated but still experienced a 51 percent rise in testosterone. Rabbits didn’t fare as well, showing mainly a 32 percent increase in undesirable DHT.1 Another study showed that giving rats tribulus protected against diabetes through antioxidant activity.2

  Animal studies can’t always be extrapolated to humans. While the research on tribulus emanating from Russia is nothing short of glowing, the herb hasn’t fared as well under Western scientific scrutiny. One study involved 15 men who got either a placebo or tribulus at a dose of 3.21 milligrams per kilogram of bodyweight daily. The subjects followed an eight-week weight-training routine. Muscle endurance increased on the bench press and leg press in the placebo group, while those in the tribulus group showed an increase only in leg press strength. No changes in body composition occurred in either group.3

  Another study examined the effects of either a placebo or 450 milligrams a day of tribulus in 22 rugby players who were engaged in weight training for five weeks. Both groups produced similar gains, along with no rise in testosterone in the tribulus group.4Still another study looked at whether tribulus raises testosterone in young men. The 21 subjects took either tribulus or a placebo for four weeks. Tribulus had no effect.5

  Given that tribulus can raise estrogen, it’s not surprising that there’s at least one case study on record of a young bodybuilder who acquired gynecomastia, or male breast formation, after using a tribulus supplement.6 DHEA can easily convert into estrogen, particularly in younger men, although it more often converts into a metabolite of DHT.

  One company is selling what it calls a “potent testosterone booster” based on the herb fenugreek. According to the company’s claims, substances in fenugreek called fenusides promote a significant rise in free, or active, testosterone. The company mentions a study in which 55 men, aged 18 to 35, all actively involved in weight training, were divided into two groups, with 29 receiving the fenugreek product and 26 getting a placebo. The subjects were evaluated three times over the eight-week course of the study. Those in the fenugreek group showed a 100 percent rise in free testosterone compared to the placebo group, along with a reduction in skin folds, pointing to lowered bodyfat.

  Sounds impressive, until you realize that this was an unpublished study done by the company making the product. No other details of the experiment were mentioned. Interestingly, fenugreek contains some of the same plant sterols found in tribulus, including protodioscin, as well as diosgenin, which was as found in Mexican yams. It contains a unique amino acid called 4-hydroxyisoleucine that has an effect similar to insulin in aiding glucose uptake. One study of cyclists showed that fenugreek seemed to increase the efficiency of muscle glycogen repletion when taken with glucose, which is precisely what insulin does.
